Near-Infrared PAT in Pressed Candy OEM: Online Blend Uniformity, Validation and Upkeep

Tablet quality has traditionally been judged by sampled testing: pull samples at intervals during compression, send them to the laboratory for assay, hardness and disintegration, and receive the result after the batch has already been compressed. The weakness is not accuracy but coverage, because deviations usually occur between two sample points. Near-infrared spectroscopy and process analytical technology move measurement onto the line, returning a result in minutes or seconds, so that whether a batch is uniform stops being an inference and becomes an observation. Making that work depends less on buying an instrument than on getting sample design, the reference method, model validation and maintenance right. In our pressed candy OEM work we treat online measurement as part of process capability rather than a laboratory tool.

Near-Infrared PAT in Pressed Candy OEM: Online Blend Uniformity, Validation and Upkeep

1. What near-infrared actually measures

NIR measures overtone and combination absorptions of hydrogen-containing groups such as C-H, O-H and N-H. It does not read an assay directly; it predicts from a mathematical relationship between spectral features and results produced by a reference method. That gives it a clear boundary. Anything correlating stably with the spectrum can be measured, including moisture, active content, blend uniformity and some polymorphic forms. Anything correlating weakly cannot, such as microbiological status, most related substances, and parameters needing destructive preparation. When assessing whether a contract manufacturer has genuine online capability, asking whether they own an instrument is the wrong question. Ask instead how many batches the model was built on, what the reference method is, and what the most recent prediction error was. Those answers separate a working tool from an expensive ornament.

2. Sample design matters more than the algorithm

Most of a model is determined by the samples behind it. The common mistake is building it from laboratory-prepared gradient samples, which look elegant on a concentration chart but carry none of the variation of real production, so the model fails the moment a raw material lot changes. The right approach samples from actual production and deliberately covers the genuine sources of variation: different raw material suppliers and lots, seasonal temperature and humidity, different blending times and speeds, different granule moisture levels, and the upper and lower limits the formulation allows. Several dozen batches are typically needed, spread evenly across the assay range so no large gaps remain. Every sample needs both a spectrum and a reference value, and the repeatability of that reference method sets the ceiling on the model. Noisy laboratory data will not yield a reliable relationship regardless of the algorithm.

3. Where to place the measurement

NIR usually sits in three places on a tablet line. At the blender discharge or hopper it monitors blend uniformity and determines end point, which beats fixing a ten-minute blend: the spectrum tells you when the mixture stops changing, avoiding both the segregation that over-blending causes and the risk of under-blending. Beside the intermediate bin or feeder it measures granule moisture and content for in-process adjustment. At the tablet discharge it tracks finished tablet content by time-slice group. Instruments come as handheld, at-line and in-line. Handheld units are flexible but depend on operator discipline, suiting model sampling and spot checks. Fixed in-line probes suit trend monitoring, but mounting position, probe fouling and material flow all affect the signal, so representativeness must be verified after installation to confirm the probe is seeing the whole batch rather than a fraction of it.

4. Validation and acceptance criteria

No model goes live unvalidated. Three things are required as a minimum. Internal cross-validation, showing correlation and error between predicted and reference values. External validation, using independent batches not involved in building the model, which is where real performance shows. And method comparison, running in parallel with the current test method to confirm the difference is acceptable. Judgement rests on correlation coefficient, root mean square error of prediction and relative deviation, with limits depending on intended use: a model used only for trend monitoring can be judged more leniently, one used for release decisions must be strict. Rules for extrapolation also matter. When a sample falls outside the modelled range the model should raise a flag rather than return a number, which becomes critical when changing raw material supplier or adjusting the formula.

5. Drift, maintenance and revalidation

NIR models drift, and that is normal rather than a fault. Lamp ageing, dust on the probe, changes in ambient temperature and humidity, a switch of raw material supplier, a shift in excipient particle size distribution all pull predictions away over time. A maintenance regime needs at least a performance check against known samples per batch or daily with results trended, defined triggers for revalidation such as repeated out-of-tolerance checks, a change of critical raw material or major equipment overhaul, and records of every maintenance action with its rationale. Plenty of plants budget well for the instrument and then, two years later, nobody maintains the model and testing slips back to sampling. That is a management failure, not a technical one. A brand can address it in the quality agreement by making model maintenance records and the annual revalidation report available for review.

6. Where PAT data fits in release

Under the current compliance framework NIR is mostly used for process control and internal control, with finished product release still resting on the statutory test method. The practical approach folds online data into the batch record and the annual review: record monitored trends and how any excursions were handled in the batch record, and use the annual product quality review to summarise process capability and correlate it with deviations, using data to demonstrate that the process is stable. This pays twice over. Internally, drift is caught earlier and rejected batches fall. Externally, when a client or auditor asks how you guarantee that every tablet carries the same amount of active, the answer is a continuous process record rather than a handful of passing test points. For a contract manufacturer that distinction, between testing quality in and building it in, is where the real difference lies.
