Capsule OEM Fill Weight Control: Dosing Tube, Piston Pump and Vacuum Filling Compared

Fill weight variation is an honest indicator. It can hardly be concealed by sorting after the fact, and a client with a balance can read a plant's capability from it in minutes. Yet when variation appears, the instinct is to adjust the machine, when the real question is whether the dosing principle suits the material. Every dosing method depends on particular powder properties, and a free-flowing powder of low bulk density needs a different approach from a poorly flowing powder that compacts readily. In our capsule OEM work we characterise the material first, select the dosing mechanism second and only then discuss parameters. Reversing that order is how plants end up tuning endlessly with little to show for it.

Capsule OEM Fill Weight Control: Dosing Tube, Piston Pump and Vacuum Filling Compared

1. Three dosing principles and what suits them

Hard capsule filling relies on three main approaches. The dosing tube dips into a powder bed, forms a plug and ejects it with a piston; accuracy depends on how consistently that plug is densified, and it suits powders and granules of moderate flow with a tight particle size distribution, making it the most versatile option. The piston pump draws a fixed volume into a chamber and discharges it, suiting finer powders and pellets with good flow, and it is sensitive to changes in bulk density. Vacuum filling pulls material into the dosing chamber under negative pressure, suiting cohesive fine powders that bridge easily, at the cost of heavier demands on vacuum stability and filter maintenance. Combination feeders exist for pellets and tablet-in-capsule formats used in modified release or fixed-dose combinations. None is universally better; each depends on different material behaviour.

2. Powder properties that decide accuracy

Three properties matter most. Flow determines whether the dosing chamber fills consistently, assessed through angle of repose, compressibility index or flow function; poor flow produces bridging, rat-holing and intermittent feed. Bulk density and its within-lot variability set the basis for volumetric dosing, so a drifting bulk density moves fill weight even when the volumetric dose is perfect. Particle size distribution and the fines fraction affect cohesion and how the plug densifies, with excess fines raising electrostatic adhesion and wall build-up. Oily or volatile-containing materials and hygroscopic extract powders change the surface state of the powder as well. Measuring these during development and setting acceptance ranges is far more effective than trialling parameters on the shop floor.

3. Set-up, from empty machine to first pieces

Set-up before routine filling should follow a fixed order. Confirm shell feeding and separation rates first, because defective shells alone produce rejections and apparent fill variation. Then set dosing parameters, including dosing tube penetration depth, powder bed height, tamping strokes and vacuum level, changing one at a time and recording the result. Then run first-piece confirmation: weigh a consecutive series of capsules, calculate mean and relative standard deviation, and check lock length and appearance alongside. Passing first pieces is not the end, because powder bed stability over time still has to be confirmed; a great deal of drift appears ten or fifteen minutes in, when the hopper no longer feeds as it did at start-up. Keep complete set-up records, which together with material lot and ambient conditions form the basis for later traceability.

4. In-process monitoring and sampling rules

Monitoring rests on frequency and sample size. The usual practice weighs samples taken at intervals, typically ten capsules or more, calculating mean and deviation and watching for individual out-of-limit units. Sampling has to cover start-up, steady running, after refilling the hopper, after a powder lot change and near the end of a run, because that is where risk concentrates. A slow downward drift in the mean usually points to a falling powder bed or changing densification rather than random machine behaviour. Modern fillers often carry online check-weighing that monitors every capsule or groups of capsules and rejects automatically; such systems need periodic verification with standard weights, and rejection records belong in the batch record.

5. Common faults and how to investigate them

High mean, low mean and high variability are three different problems. A systematically low mean usually means insufficient densification, an incompletely filled dosing chamber or a powder bed that is too low; a high mean usually indicates over-tamping or over-sized dosing settings. Variability needs to be split into random and periodic. Random variation usually comes from inconsistent flow or static adhesion; periodic variation usually follows machine stations, such as a worn dosing chamber or one station that feeds poorly. One easily missed case is worth separating: wide variation in empty shell weight makes total weight look unstable when the fill is actually fine, so weighing shells and contents separately localises it. Investigate along four lines, material, parameters, equipment condition and shells, rather than judging them together.

6. Improving the material instead of the machine

When the material does not suit the existing mechanism, changing the formulation or pre-treatment usually beats forcing the machine. Granulation raises flow and bulk density so volumetric dosing becomes stable. A glidant reduces cohesion, though the dose must be controlled because excess lowers bulk density and can delay disintegration. Controlling the fines fraction and moisture range reduces wall build-up and static. For hygroscopic materials, controlling room relative humidity and exposure time matters. These changes interact with dissolution and disintegration, so they must be validated together during development rather than tuned purely to make weighing look good. The sensible route settles formulation, process and equipment in a single technical assessment.
