Enteric Release in Capsule OEM: Coating Materials, Curves and Process Windows

In technical discussions with a capsule OEM supplier, enteric delivery is one of the most frequently requested capabilities and one of the most consistently underestimated. Brand owners usually describe the goal simply: let the active bypass the stomach and release in the intestine. In production terms, though, enteric delivery is a coating engineering task with explicit performance targets. The capsule must resist simulated gastric fluid for a defined period, then release at a designed rate once it reaches a particular intestinal pH, and it must keep doing both throughout its shelf life. Miss any one of those and the product only looks enteric. This article covers coating material selection, coater parameters, release testing and the scale-up risks that decide whether a programme is genuinely deliverable.

Enteric Release in Capsule OEM: Coating Materials, Curves and Process Windows

1. Which performance targets enteric delivery actually implies

Translated into measurable terms, enteric delivery involves four requirements. The first is acid resistance: no disintegration or leakage in simulated gastric fluid for the specified duration, which is the minimum entry ticket. The second is the pH at which release begins. The pH differs markedly between intestinal segments, so a coating's dissolution threshold determines where the active is released, and choosing the wrong location is worse than not coating at all. The third is release rate: how much is released over what time once the intestinal environment is reached, which has to match the intended formulation behaviour. The fourth is stability, meaning the film must not lose acid resistance through moisture uptake, plasticiser migration or shell ageing during storage. That last point is the most neglected and is very often the reason a shelf-life failure appears.

2. Selecting the coating material

Two families dominate: acrylic resins and cellulose esters. Acrylic resins film well and their dissolution pH can be tuned through monomer ratio, which is why they are the most widely used option, but the film tends to be brittle and needs a plasticiser. Cellulose esters such as hypromellose acetate succinate offer strong acid resistance and a flexible film, yet are more sensitive to humidity and demand tighter storage conditions. Three factors drive the choice: the pH of the intended release site, whether the fill is moisture sensitive, and whether the shell is gelatin or plant based. Gelatin shells become brittle as they lose moisture in a dry environment and can deform under the repeated heat and humidity of coating, whereas plant shells tolerate this better but present a different surface energy, so the coating formulation has to be adjusted accordingly.

3. Setting the coating process parameters

Coating is fundamentally about producing a uniform, continuous film. Four parameter groups matter. Inlet air temperature and volume control solvent evaporation; evaporate too fast and the film cannot level before it sets, giving a rough surface or even cracks. Spray rate and atomisation pressure set droplet size and spread; too high a rate causes local pooling and capsule agglomeration. Drum speed and gun distance govern uniformity, and a slow drum leaves capsules insufficiently tumbled so weight gain spreads unevenly. Weight gain itself determines acid resistance directly. In practice the sensible route is a small-scale weight gain ladder study, plotting acid resistance time and release profile against weight gain, followed by selection of the production target rather than copying a supplier's recommended figure.

4. Making the release profile trustworthy

Release testing runs in stages that mimic the gastrointestinal environment: sampling in acid medium for the specified period to confirm minimal release, then transferring to intestinal pH medium with timed sampling. The number and spacing of sampling points should match the intended behaviour, and a targeted delayed or sustained product needs denser sampling. The analytical method itself requires validation, and where the fill contains several actives, their individual dissolution behaviour must be checked for consistency. There is one further criterion that is routinely overlooked: both intra-batch and inter-batch consistency matter. If six capsules from the same batch scatter widely, coating uniformity is poor, and a tidy mean curve cannot be taken as evidence of a controlled process.

5. Scale-up and shared-line production

Moving from a laboratory coating pan to production equipment, heat and airflow distribution are what most often fall out of control. In a larger drum the residence time distribution across the bed widens, so capsules near the spray gun receive more liquid and those at the periphery receive less. The result is a broader weight gain distribution and a weak point in acid resistance. Scale-up is commonly done by matching spray volume per unit surface area, and weight gain distribution rather than mean weight gain should drive release. On shared lines, enteric product should not be scheduled directly after ordinary coated product, because residual standard coating material contaminates the enteric film and undermines acid resistance, so cleaning validation must be designed specifically for that risk. Coated capsules are also humidity sensitive in storage, which calls for a higher packaging barrier.

6. Judging whether an enteric programme is credible

A brand owner can assess a factory's enteric capability by asking for three things. First, measured acid resistance and release data across multiple batches, not a single run, because single-batch data carries high risk. Second, records of weight gain distribution and intra-batch consistency, which reflect process control rather than formulation. Third, stability data showing acid resistance is maintained under accelerated conditions. If the only material available is one coating supplier's datasheet and one curve, the programme is still at formulation stage and some distance from manufacturable. Written into the technical agreement, these three items turn enteric delivery from a concept into something a supplier can actually be held to.
